work okayness, personal awesomeness
Jul. 6th, 2010 06:45 pmI had a research setback today -- one of my lines that's supposed to segregate me triple mutants seems to have had a mis-genotyped parent, so they aren't carrying one of the knockout alleles at all. I don't have any other seed that will get me this combination, so I'm going to have to make crosses again when the double mutants are ready. On the bright side, I did expect this triple to be dead, so it's not really mission-critical to confirm that immediately. Unfortunately I've also been a bit of a bonehead today, screwing up a PCR three times. (Okay, I admit there were five recipes involved, but usually I can do that.)
In general, the research is going well, though. I have lots and lots of plants that need genotyping, but in a great stroke of luck we also have undergrads who need to be trained to genotype things, and other undergrads who are well able to train them if I just point them at the plants. Clearly they will then need practice as well, so this shouldn't be totally unmanageable. And some of the mutants do seem to have a phenotype, though not a very strong one, which explains why it hasn't been noticed. I just need to work on my cloning, which will let me make the other end of the argument as to my genes' function. Work, Kylee, work!
I was extremely productive over the weekend, though. I think I'm making up for the frustrated stage of being sick, when I didn't have the energy to actually do anything useful but did have enough brain to be annoyed that I couldn't. The kitchen is very clean, except for the floor which I kind of forgot about. :) And today I ran errands. Some might say that the Molly Moon's stop was not strictly necessary, but to them I say: ginger ice cream in a waffle cone, people. Sadly,
marzipan_pig rejected my Ultimate Efficiency Plan for tonight, in which I was able to paint loft-bed pieces while she quilted, but relaxing and working on my knitted Dalek will be a good thing too.
